To test this antibody, I stained PBMCs for 30 minutes at room temperature in the dark at serial dilutions of 1:25, 1:50, 1:100, 1:200, and 1:400.

As you can see from the stainings CD69 is not well expressed in PBMC, and would have been better to test in tissue cells. Nevertheless I would go ahead and stain at a 1:100 dilution.
