To study the proliferation of cells by flow cytometry.

Though protocol recommends the concentration of 10uM, this works good at lower concentration as well.

Results Summary

The result here are shown for quantified proliferation of intestinal organoids. I used two different culture condition for organoid and estimated the proliferation by BrdU incorporation. The concentration of the BrdU used here is 5uM for 0.2million cells and incubated for 45 mins at 37 degree celsius. After BrdU labeling cells were stained with viability dye for 20 mins in ice and treated with DNase I for 1 hr covered with aluminium foil. After DNase I treatment cells were kept in BrdU staining buffer for 15 mins and washed twice before staining with anti-BrdU PE antibody for 30 mins on ice. The cells were finally acquired on BD Symphony A3.

Very good staining and proliferation result.

Long protocol and expensive.

The Bottom Line

Because it can combine with multiple parameter, proliferation of different population of cells can be estimated simultaneously. So, I would definitely used it.
