Zombie UV fixable viability kit

I used this viability kit for my invitro proliferation assays for organoids and immune phenotyping of my tumor microenvironment assays. It is a perfect viability kit that discriminates dead cell populations from live cells.

This is light sensitive dye so always prepare fresh or 1-2 hours before use.

Results Summary

The following result is obtained from the intestinal organoid culture proliferation experiment. I stained 0.2 million cells harvested from organoids with Zombie UV at 1:1000 dilution for 20 mins on ice and then fixed with 2% of paraformaldehyde for 30 mins on ice. The zombie UV stained and discriminated against the dead population from the culture. The dye is excited by a UV laser and emits in the DAPI channel. So dead population is shown here as DAPI positive contour.

Summary

The Good

Works very good even at lower concentration. I used it between 1:500 to 1:1000 dilutions.

The Bad

Light sensitive but it's not a drawback. Use Aluminum foil.

The Bottom Line

The Zombie UV dye is excited by a UV laser and emits in the DAPI channel. So it leaves other important channels open. In complex immune phenotyping experiments this could be very helpful.
