The goal of my experiment is, to find how different cell treatments influence the flipping of calreticulin to the exterior cell membrane. As one of three correlates of immunogenic cell death.

Primary Incubation

0.1uL Zombie Green viability stain in 100uL PBS, incubated at room temp for 15 mins.

Blocking Agent

Human TruStain Fc block, 5uL diluted in 95uL FACS buffer (5% FBS in DPBS), incubated at 4C for 10 mins.

Secondary Incubation

0.5uL of Calreticulin antibody diluted in 100uL of FACS buffer (5% FBS in DPBS), incubated at 4C for 20 mins.

Flow cytometry in a beckman coulter cytoflex cytometer.

Results Summary

My data showed increased calreticulin cell surface expression with certain methods of cell killing (such as gamma irradiation of cells), in a dose-dependent manner.

Summary

The Good

Works better than other Calreticulin antibodies.

The Bad

Have had a lot of difficulty getting consistency with adherent cells.

The Bottom Line

It's a great antibody for a fairly difficult test when it works, and it has worked better than the abcam ones we have tried in this lab. But, calreticulin staining is very finicky in general and this antibody is no exception.
