Product Name:

Amplex™ Red Cholesterol Assay Kit

Catalog Number:

A12216

Info: Supplier Page

I am interested in measuring membrane cholesterol upon statin treatment from mammalian cells. I used the Amplex™ Red Cholesterol Assay Kit for measuring total cholesterol from membrane preparations.

Experimental Design and Results Summary

Application

ELISA

Starting Material

Cell lysates or membrane

Tips

Can avoid cholesterol esterase to get non-esterified membrane-bound cholesterol.

Results Summary

Membrane cholesterol content of CHO-K1 cells treated with increasing concentrations of lovastatin. Values are expressed as absolute cholesterol content in cell membranes and are normalized to protein content. Data represent means ± SEM of three independent experiments (* and *** correspond to a significant difference [P < 0.05 and P < 0.001] in membrane cholesterol content in lovastatin-treated cells relative to untreated cells). The inset shows cholesterol/phospholipid ratio (C/P, mol/mol) under these conditions. Data represent means ± SEM of three independent experiments (** and *** correspond to a significant difference [P < 0.01 and P < 0.001] in C/P ratio in lovastatin-treated cells relative to untreated cells).

Summary

The Good

Rapid assay, provides both free and esterified cholesterol, and has got a good dynamic range.

The Bad

None.

The Bottom Line

This is an easy to use kit for rapid measurements of cholesterol content.
