I am interested in understanding the mechanisms of oncogenic WNT signal transduction. I use precise genome editing by CRISPR/Cas9 and ssODN to generate endogenous mutations in the components of the WNT pathway to understand their role in the WNT signal transduction. This method requires Cas9 protein and so in this experiment, I purified Cas9 protein from E. coli, and the purity was determined by staining the gel with the GelCode Blue reagent.

Wash gel with water for 10 min. Repeat two more times before adding the reagent.

Results Summary

I get good staining if the gel is pre-washed with ultra pure water for three times. They typically stain for 1-3 hours followed by several washes of ultra pure water.
