Company:

Proteintech

Product Name:

mCherry Polyclonal Antibody

Catalog Number:

26765-1-AP

Clone Number:

N/A

This polyclonal antibody from Proteintech is suitable for the detection of mCherry and mCherry-conjugates via Western blot as well as IP (other applications were not tested). Although optimization between different samples is necessary, detection is easy and specific and the results are more than satisfying.

Experimental Design and Results Summary

Applications

Western blot

Sample

Human cancer cell lines (whole cell lysates) e.g. HEK293, MCF-7

Primary Incubation

1:1000 at 4°C overnight

Blocking Agent

5 % BSA in TBST

Secondary Incubation

1:2000, 60 min at RT

Tertiary Incubation

None

Detection

HRP, Chemiluminescence (West Dura Extended Duration Substrate) in Elysia-raytest detector (3 min exposure)

Results Summary

Detection yielded a specific band in the case of free mCherry at the size of approx. 27 kDa. Exposure times were low with a maximum of a couple of minutes. But, also mCherry fusion proteins are easily detectable. Moreover, the pull down of such proteins was done using this antibody and in principle works but needed to be optimized for IP.

Additional Notes

Antibody working solution can be stored at +4°C for up to two weeks without any quality loss. For optimization, vary exposure times/antibody dilution e.g. 1:500

Summary

The Good

Works well for WB and IP, no unspecific bands.

The Bad

Optimization for different samples needed.

The Bottom Line

Polyclonal antibody 26765-1-AP from Proteintech is a decent antibody for use in Western blot and IP, while other applications were not tested but are also possible. All in all, I can warmly recommend it for detection of mCherry or mCherry fusion proteins.
