Extracellular vesicles (EV) were harvested from 2D cultures of stem cells and various types of cardiac cells. The miRNeasy Micro Kit was used to lyse EVs and isolate miRNA for sequencing and comparison of miRNA cargo between different EV types.

For EV miRNA, use the minimum required volume for elution to ensure sufficient concentration for downstream analyses.

Results Summary

Total RNA was successfully isolated from EVs and miRNA was specifically enriched using this kit. Total RNA was of measurable quantity and of sufficient quality when measured with a nanodrop. Enriched miRNA was also QC'd using a bioanalyzer before library preparation and sequencing, which revealed numerous significant differentially expressed miRNAs based on cell type.

Summary

The Good

Intuitive kit design, slightly lower price than some higher end kits, sufficient for miRNA sequencing.

The Bad

Slightly lower yield and purity of isolated RNA than the most expensive kits out there, still requires phase separation step unlike some higher end column-based isolation kits.

The Bottom Line

Great kit for efficient and effective miRNA isolation from higher number of samples. Great as a first kit when yield and purity aren't major hurdles with samples of interest.
