Claudin-5 Antibody [Alexa Fluor® 647]

Catalog Number:

NBP2-71329AF647

Clone Number:

OTI1G4

Claudin-5 expression on brain endothelial cells from tamoxifen-inducible Claudin-5 knock-out mice was measured by flow cytometry.

Experimental Design and Results Summary

Applications

Flow cytometry

Sample

Mouse brain endothelial cells

Primary Incubation

1:100, 30 minutes on ice

Blocking Agent

TruStain FcX™ (anti-mouse CD16/32) Antibody

Secondary Incubation

N/A

Tertiary Incubation

N/A

Detection

Cytoflex S flow cytometer (Beckman Coulter)

Results Summary

We expected that close to 100% of brain endothelial cells would be positive for Claudin-5, but only 2% of cells were positive for this Claudin-5 antibody. The signal was bright, but the antibody does not seem to work well for flow cytometry.

Summary

The Good

Bright signal.

The Bad

Poor affinity for Claudin-5 in fixed primary brain endothelial cells.

The Bottom Line

This antibody does not work that great for flow cytometry.
