We equipped the OU lab with the Lonza 4D Nucleofector System for the electroporation of different cell types. The system requires dedicated consumables and we tested different vessel sizes. These cuvettes can host large amounts of cells (up to 10M) in a relatively small volume of 100 ul and reactive protein/RNA complexes or plasmid DNA. The kit is compatible with a large list of primary immune cells, both from humans and mice. We validated the kit for primary lymphocytes T (human/mouse) and lymphocyte B-derived cell lines.

Results Summary

Excellent results in matter of transfection efficiency and and cell viability. The small volume and the high concentration of the cells allow to save expensive reactives, such as Cas9 protein and RNAs. The large amount of efficiently electroplated cells per run is sufficient to perform complete experiment with several conditions.

The Bottom Line

Excellent results for primary cells DNA or protein transfer with high viability. The large number of cells processed at once allows to reduce the number of electroporations required to fill all the conditions of the experiment. The fixed programs and kit grant easy manipulation and reproducibility.
