We are developing chimeric, secreted proteins that require mammal/human post-translational modifications. We, therefore, used the suspension HEK293/F line. After several attempts with cheaper media (DMEM, low serum, or IgG depleted), we decided to move on with the more expensive 293 SFM II. No regrets at all! The cost is largely compensated by the much higher yield of protein produced by the cells maintained in suspension.

Cells can grow in a very high density, better if replenished with glutamine at day 7.

Results Summary

We achieved very high quantity of secreted protein by this cell line. The cells maintain good viability even after 10 days of culture in the same medium (67% viability, 2M cells/ml), provided with a small supplement of glutamine at day 7.
