Venetoclax from Sellekchem is a potent Bcl-2 inhibitor. We regularly use it for studying the Bcl-2 protein family network and apoptosis. However, at higher concentrations, Venetoclax most likely exerts off-target effects.

(Human) Cancer cell lines.

Tips

Aliquot upon arrival and store (e.g. 100x working concentration at -20°C). For efficient and specific inhibition of Bcl-2 we use ABT-199 at 5 µM. At about 15 µM we observed off-target effects. Determining of IC50 initially, is recommended.

Results Summary

Venetoclax specifically and potently inhibits Bcl-2 protein resulting in apoptosis execution in Bcl-2 dependent cell lines such as many CML and AML cells. Also in Sarcoma cell lines we revealed good results in combination with proteasome inhibition.
