My research involves studying compartmentalized dopaminergic signaling. We grow 3-dimensional polarized cell cultures on transwell membranes to observe apical and basolateral localization of proteins of interest.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

Madin-Darby Canine Kidney (MDCK) cell line

Primary Incubation

1:500 dilution, 1hr, RT

Blocking Agent

2.5% BSA in 0.1% TBS-Triton

Secondary Incubation

1:1000 dilution, 20 min RT

Tertiary Incubation

N/A

Detection

Fluorescence microscope

Results Summary

Staining looks very clean; however ZO-1 is supposed to help verify polarized cultures as a tight junction protein and we saw this staining on cells that still looked quite flat, not polarized.

Summary

The Good

Could see clear staining.

The Bad

May be more of a membrane marker, not for polarized cultures.

The Bottom Line

Decent product, may not be useful for those who want to verify that 3-dimensional cultures polarized into developing apical and basolateral membranes.
