This ProLong mounting medium with DAPI is an easy-to-use mounting medium for fixed microscopy samples. I regularly use it for my fluorescence/confocal microscopy samples (fixed cells) which gives a nice nuclei staining which remained stable for several days and preserves the rest of the sample well for many weeks.

Fixed cells (human cell lines) on coverslips

Tips

In some cases the DAPI staining remained only stable for a week or two. Samples should be imaged asap. 5 µl of medium is enough for a 12 mm coverslip.

Results Summary

After mounting samples, the mounting medium usually solidifies well over night. The staining is stable for at least a couple of days. However, background is possibly higher than for mounting media without DAPI.

For everyone who wants to have an easy-to-use DAPI staining combined with the mounting step I can recommend to try this product. However, the combination of these steps might have draw-backs in staining stability and background staining as mentioned above.
