DMEM/F-12 is a balanced mixture of DMEM and Ham's F-12 that allows reliability, consistency, and improved control in mammalian cell culture. It requires supplementation of FBS. We usually buy the media with L-glutamine already. For most cells, I could easily substitute the DMEM and F12 as they are very similar in composition.

Don't leave the media overtime at room temperature or open to avoid pH change that may that affect cell growth.
In our lab, we do use Gibco DMEM/F-12 for different mammalian cell lines and could easily substitute the DMEM. We usually buy the media with L-glutamine already and add FBS sterile before use it.
