NRF1 protein in human mesenchymal stem cells (hMSCs) was detected using an antibody-based approach. To prepare the antibody, it was diluted to a concentration of 1:5000 in a solution containing 5% BSA and TBST (Tris-base buffered saline with tween-20). The diluted antibody was then incubated with the hMSCs overnight at 4 ℃. The hMSCs were subsequently exposed to a secondary antibody (rabbit, 1:10000) for 1 hour at room temperature. Finally, the signal was amplified using HRP and detected by exposing the cells to a chemiluminescent substrate for 20 seconds.

NRF1 antibody from abcam (ab175932) is showing highly competitive to Westen blot analysis in hMSCs.NRF1 prtotein (total protein: 20 μg)has been detected in 1:5000 dilution. First lane cell lysate was isolated from the control MSCs with 24 hr serum starvtation and second lane cell lysate was prepared from the MSCs in 10% FBS containing medium.
