I used in to detect PTX3 in human plasma samples. The plasmas were separated with density gradient medium during the PBMC isolation and were stored in -80 Celsius.

I diluted my samples as 100uL plasma + 50uL dilution buffer.
The starting range was a bit high than we wanted. If it would be 35pg/mL that would be better. Also, the background was quite high, so we had to optimize to get rid of it, we used 150uL for each sample and diluted everything with 50uL dilution buffer.

High background.
I recommend it for human plasma samples. The assay can also be used on human plasma samples that were stored in -80 for long periods.
