Antibodies are produced by plasma cells (PC) that are terminally differentiated B cells and play crucial roles in host protection against infections but are considered to mediate autoimmunity in patients with rheumatic diseases, such as systemic lupus (SLE) or rheumatoid arthritis (RA). Our research aims at understanding the biology of B cells and antibody-secreting cells (ASC) in health and disease, but in this experiment we are looking for CD4 T cells.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Mouse WT splenocytes

Primary Incubation

Surface staining with CD4 PE-Cy7 (1/200), 20 min at 4C

Blocking Agent

Fc block (2.4G2 clone)

Secondary Incubation

N/A

Tertiary Incubation

N/A

Detection

Flow cytometer

Results Summary

We gated lymphocytes and singlets. Within the singlets gate we can find B cells (CD19+) that are the majority of the lymphocytes in the spleen and CD4+ T cells, that represent around 20% of the lymphocytes in the spleen.
