In our lab, we use this kit for Mycoplasma detection on cells supernatant. We do follow the protocol as it is presented (no optimization needed). On both fresh collected and frozen samples, -80C results are reliable. Also it allows to detect mycoplasma on other samples like medium, fbs, pbs, trypsin with a pre-dilution step (1:10) with buffer or sterile distilled water. Results have been always reliable.

To avoid false positives related to the presence of cells in the supernatant, do a centrifuge step collecting at least 2ml of supernatant; then keep the supernatant to test and discard any pellet present;

The assay buffer or distilled water also serves as a negative control. We purchased separately extra assay buffer. We bought also the Positive control for MycoAlert® PLUS Mycoplasma Detection Kit which includes a lyophilized positive control and assay buffer for reconstitution.
