This stripping buffer is the perfect choice if you have sensitive antibodies. Stripping is working quite well in gerneral (15 min, RT) - sometimes not thoroughly enough though. Always depends on the antibodies used and incubation time.

Western Blot - antibody stripping

Starting Material

Nitrocellulose membrane loaded with primary/secondary antibodies for Chemiluminescence detection (HRP-coupled).

Tips

We use the same buffer for up to 4 times without any quality loss. For larger amount of antibody increase incubation time.

Results Summary

Strips off primary and secondary antibodies bound to the Nitrocellulose membrane. Antibodies are usually removed reliably, so that after washing and re-blocking a new round of antibodies can be used with the same membrane. When stripping was insufficient, you might detect (traces of) the antibodies used before. Unfortunately, we observed that for stripping of larger amount of antibody the buffer does not work within the recommended 15 min at RT.

We use this stripping buffer for all our Western Blot with satisfying results, although optimization was needed for every new antibody tested.
