Antibodies are produced by plasma cells (PC) that are terminally differentiated B cells and play crucial roles in host protection against infections but are considered to mediate autoimmunity in patients with rheumatic diseases, such as systemic lupus (SLE) or rheumatoid arthritis (RA). Our research aims at understanding the biology of antibody secreting cells (ASC) and B cells in health and disease. Here, we were looking at IgA+ PCs in the spleen of a WT mouse.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

C57BL/6 spleen

Primary Incubation

surface staining with IgA FITC (1:100), 20 min at 4C

Blocking Agent

Fc block (2.4G2 clone)

Secondary Incubation

Detection

Flow cytometer BD CANTO

Results Summary

We could detect IgA expressing plasma cells. Around 20% of the plasma cells (CD138hi) in the spleen. The signal is not very strong, but a populations of IgA+ cells can clearly be identified. To improve the staining the antibody can be used at a 1/50 dilution.

Additional Notes

The anti-IgA APC (same company) works better.

Summary

The Good

It works.

The Bad

Weak signal.

The Bottom Line

The signal is weak, so if you can is better to use anti-IgA APC (same company).
