Antibodies are produced by plasma cells (PC) that are terminally differentiated B cells and play crucial roles in host protection against infections but are considered to mediate autoimmunity in patients with rheumatic diseases, such as systemic lupus (SLE) or rheumatoid arthritis (RA). Our research aims at understanding the biology of B cells and antibody-secreting cells (ASC) in health and disease. Although, we are usually not staining T cells, or just staining them for exclusion, in this case we stained CD3 T cells in spleen and mLN of WT mice.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Spleen and mLN of C57BL/6

Primary Incubation

Surface staining with CD3 APC-Cy7 (1:200), 20 min at 4C

Blocking Agent

Fc block (2.4G2 clone)

Secondary Incubation

N/A

Tertiary Incubation

N/A

Detection

Flow cytometer with a red laser (in this case a CANTO)

Results Summary

We stained CD3 T cells in spleen and mLN, as well as CD19 B cells. First, we gated in lymphocytes and singlets. Regarding frequencies of T cells, as expected the frequency of CD3 cells in higher in the LN than in the spleen.
