In different healthy and pathological contexts, CD4 and CD8 T cells differentiate into a range of specialized effector and regulatory subsets. In mouse models of aging and autoimmune diseases we study the effector and memory phenotype of CD4 and CD8 T cells. We identified different subsets of naïve and activated CD4 and CD8 T cells with an anti-Ly108 BV711 antibody in 24-month-old C57BL/6 mouse.

Primary Incubation

Washed twice in PBS and incubated with L/D fixable aqua dye 20 min at 4°C, then washed twice in PBS.

Surface staining with anti-CD3 BUV805, anti-CD4 BUV495, anti-CD8 FITC, anti-CD62L BV605, anti-Ly108-BV711 1/150 etc., in Cell Staining Buffer (Biolegend) incubated 30 min at 4°C.

Results Summary

Different populations of CD4 and CD8 T cells are identified: CD62L+Ly108- naive cells, and CD62L-Ly108+ activated cells.

Not as bright as other fluorochromes, careful compensations controls should be done.

The Bottom Line

Usable, and good option to use the BV711 channel. Some caution for the compensations should be taken, in particular in PerCP Cy5.5 channel.
