In different healthy and pathological contexts, CD4 and CD8 T cells differentiate into a range of specialized effector and regulatory subsets. In mouse models of aging and autoimmune diseases we study the effector and memory phenotype of CD4 and CD8 T cells. We identified memory and naive CD4 T cells with an anti-CD62L BV605 antibody in pre-lupus NXBxW F1 mice.

Primary Incubation

Washed twice in PBS and incubated with L/D fixable aqua dye 20 min at 4°C, then washed twice in PBS.

Surface staining with anti-CD3 BUV805, anti-CD4 BUV495, anti-CD8 FITC, CD44 BV785, anti-CD62L BV605 1/300, etc., in Cell Staining Buffer (Biolegend) incubated 30 min at 4°C.

Detection

Violet laser on Symphony cytometer.

Results Summary

We could identify CD62L+CD44- naive, CD62L-CD44+ effector, and CD62L+CD44+ central memory CD4 T cells in NZBxW F1 mice.
