Our group is interested in using TCRs for therapeutic purposes. As such, for our experiments we commonly transduced TCR constructs or other proteins to primary cells and cell lines. Retronectin is our regent of choice to increase our transduction efficiency.
Retroviral Transduction
Primary PBMCs or cell lines
For primary cells and precious material we use the same aliquot of retronectin twice. For cell lines we store the used retronectin in -20 and that/use it 6+ times.
The transduction efficiency using retroviruses as the delivery method is much higher when retronectin is used.

Easy to use, works well.
Very expensive.
The regent works well and is reliable between batches. Definitely worth using when the material is precious and the highest transduction efficiency is required. It is however very expensive.
