I am studying the effect of extracellular phospholipid on microglia signalling cascade. In response to the phospholipid, there is an increase in phosphorylation of Akt at Serine 473 in primary murine microglia. Changes in the expression of phospho Akt were detected using immunoblotting.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Primary murine microglia

Primary Incubation

1:1000, 4 degree C, overnight

Blocking Agent

5% milk

Secondary Incubation

1:5000 anti rabbit

Tertiary Incubation

N/A

Detection

Horseradish peroxidase (HRP) substrate

Results Summary

The antibody was very sensitive and gave a single specific bands. The bands indicated increased expression of phospho AKT in reponse to the extracellular phospholipid in primary murine microglia. This antibody is used to investigate AKT pathway.
