I use this buffer to burst red blood cells when I process mouse cell tissues in order to obtain lymphocytes for future analysis.I harvest splenocytes, bone marrow cells or liver cells and I use 1.5mL of the ACK lysis buffer per spleen, BM or liver, incubate for 1min and then stop the reaction with 13mLs of 10x PBS.

Results Summary

After processing each of the tissues or the blood with this buffer, my cell pellets look white indicating successful ACK lysis. This makes future FACS analysis more effective.

If the cells are left in the buffer longer than 1 minute, you risk killing some of your cells of interest.
