The acetylation on the Lysine 27 of the histone H3 is a marker of open and active chromatin. This antibody was used to evaluate the chromatin state and establish the epigenetic landscape in different tissues (kidney, adrenal gland).

Experimental Design and Results Summary

Applications

ChIP

Sample

Dissociated, crosslinked tissues ( adrenal gland, kidney)

Primary Incubation

1% formaldehyde for 10' at RT (crosslink)

Blocking Agent

Glycin 0,1M final concentration (quenching)

Secondary Incubation

1ug antibody for 5ug sonicated chormatin, ON at 4°C rocking

Tertiary Incubation

Protein G coated beads, 30ul/ug of antibody, 2 hours at 4°C rocking

Detection

qPCR on precipitated chromatin or sequencing

Results Summary

The antibody reliably defined a precise pattern of histone H3K27 acetylation in different cell types and tissues. The significant enrichment of the precipitated regions showed a narrower distribution compared with the H3K4me3, suggesting a better resolution of the open/active chromatine regulatory regions.
