We are isolating trophoblasts from placental tissues, and the prefinal step involves the separation of trophoblasts from stromal cells. The columns allow for negative selection using magnetic beads conjugated to stromal antibodies, but not all of them are filtered through.

Prevent bubbles from entering the column. Clogs may occur if suspension has contaminants that may block the path (e.g., blood clots, debris aggregates etc). May push down with plunger to increase filtration speed.

Results Summary

Despite negative selection, we still get contaminating stromal cells after the initial run through. This is not significant, although other factors such as the antibody used may affect isolation as well.
