A mouse model with I4895T mutation in type1 ryanodine receptor displays muscle weakness and atrophy. Muscle of this mutant model showed decreased SR Ca2+ transient, decreased cytosolic Ca2+, increased mitochondrial ROS, increased ubiquitination, and increased ER stress compared to WT.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Skeletal muscle of I4895T and WT

Primary Incubation

1:500 for overnight

Blocking Agent

Bio-Rad EveryBlot Blocking buffer

Secondary Incubation

1:10,000, 1 hour at room temperature (StarBright 700)

Tertiary Incubation

No tertiary incubation.

Detection

Bio-Rad ChemiDoc MP Imaging System

Results Summary

Ab detects total ubiquitination in the muscle lysates. I4895T of RyR1 mutation greatly increases the total ubiquitination.
