We use TrueView to reduce autofluorescence in brain, liver and heart tissues. TrueView (mounting media with DAPI) was added after all antibodies were applied to the tissue. The staining becomes clearer afterward, usually time of exposure is 3-5 min. The image represents one brain area of mouse tissue with or without TrueView (green staining only represents background intensity).

The Bottom Line

In the periphery of some tissues the DAPI staining can be quenched. Also, when using TrueView without DAPI counterstaining (the product that does not includes DAPI in the mounting media), DAPI staining will fade with time.
