We are interested to study tissue-resident macrophages in various tissues. As a positive control for CD18, we stained the mouse spleen.
Immunofluorescence
PLP fixed 14 µm cryosections of mouse spleen from a 10 week old C57BL/6J mouse
1:200 overnight at room temperature
1% BSA, 1% FCS, 0.3% Triton X, 2% mouse serum, 2% goat serum
1:500 goat anti-rat alexa fluor 555 1h at room temperature.

Images were acquired using an inverted Zeiss LSM780 confocal microscope with an 20x 0.8NA air objective.
Very nice staining of CD18+ cells in the spleen.
