The directions in the manual provide example dilutions for the biotinylated antibody, HRP-streptividin, and Assay Diluent B. This helps tremendously as normally I need to spend extra time determining the best dilution factors. They also provide extra assay diluent A for samples that need to be diluted 100-500 fold. I will continue using this brand due to the ease of operation and dilution factors.

Ensure that you have the correct dilution factor for serum; it is not the same as plasma because platelets store BDNF.

Results Summary

The first plates ran with plasma BDNF at a 1:10 dilution factor yielded nearly perfect results. The 'sweet spot' would be a 1:20 factor to fall within the curve. For serum BDNF, the dilution factor of 1:10 was not enough, plates returned highly saturated and a suggested 1:100 factor was used. I will update when we have received these kits and rerun the experience with higher dilution.

The Bottom Line

This kit works and our standard curves always return correctly and comparable to the QC of the product. Just ensure you use the correct dilution factor for serum and plasma samples as this will yield either too saturated or too little optical density. Again, that is 1:20 for plasma BDNF, and 1:100 for serum BDNF.
