Company:

Abcam

Product Name:

Caldesmon

Catalog Number:

Ab32330

Clone Number:

Actin binding protein that is used to label smooth muscle cells in a culture or tissue. The antibody is useful in labeling total Caldesmon. However, 2 different isoforms of the protein occur and the antibody does not differentiate between the two isoforms on a tissue section. However, the same antibody can label the the two isoforms when run on western blot due to difference in the size of the two isoform.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

Murine aortic tissue section and homogenate

Primary Incubation

1:100 dilution, O/N at 4c

Blocking Agent

5% BSA

Secondary Incubation

Anti rabbit secondary

Tertiary Incubation

1:200, 45 mins at room temp

Detection

Using BioRAD chemidoc for western and fluoresence microscope

Results Summary

Combine staining analysis with western blotting data helps to easily differentiate between the low molecular weight and high molecular weight isoforms of the protein and workout the ratio of expression of the each isoform to the total Caldesmon expression. It is a nice antibody considering that there are not many antibodies out there that are specific to synthetic smooth muscle cells.

Summary

The Good

Can be used in immunofluoresnce and westernblotting

The Bad

Can not differentiate between 2 isoforms in immunofluorescence study

The Bottom Line

Combine staining analysis with western blotting data to provide definitive, conclusive result. Overall a good antibody considering there are not many definitive antibody that label synthetic smooth muscle cells.
