Mito tracker red is a far red-fluorescent dye that stains mitochondria in live cells and can be used for mitochondrial localization. The dye is well-retained after aldehyde fixation and even subsequent permeabilization with detergents.

To label mitochondria, cells are simply incubated with MitoTracker probes, which passivelydiffuse across the plasma membrane and accumulate in active mitochondria. Once theirmitochondria are labeled, the cells can be treated with an aldehyde-based fixative for samplesthat need fixation to allow further processing of the sample.
