I have used this cell proliferation kit to quantify the cell cytotoxicity of drugs/compounds in vitro for the downstream assays. Human cells seeded in 96 well plates were treated with different drug concentrations and with vehicle control. After 24 hours of the treatment, the cell proliferation reagent WST-1 was directly added 20 ul/ well in the media. After 4h hours of incubation at 37 °C, the multiwell plate was scanned with a scanning multiwell spectrophotometer (ELISA reader) to quantify the formazan dye formed, which is indirectly proportional to the cell cytotoxicity.

The dye solution can be heated at 37 °C, if it precipitates.
The gradient of yellow color was observed with different drug concentrations and a decrease in the absorbance was measured with an increase in drug concentration.
Can directly add to the wells without any washing.
Dye solution may precipitate with long storage.
