Our research aims at understanding how some CD4 T cells become autoreactive in melanoma patients treated with check point inhibitors. CD4 T cells are usually identified as CD3+ CD4+ cells. In this experiment, we used Nivolumab to detect PD-1 positive CD4 T cells.

PMA/ionomycin/Calcium-stimulated healthy donor's PBMCs

Primary Incubation

Surface staining with Nivolumab at 1/1000 (20 min in the dark at 4C)

Blocking Agent

none

Secondary Incubation

Surface staining with anti-CD4 PEDazzle594, anti-CD3 BUV495, etc, and PE-conjugated anti-human IgG4 secondary antibody to detect Nivolumab (20 min in the dark at 4C)

Tertiary Incubation

Cells were fixed and permeabilized with a commercial kit to counterstain for cytokines.

Yellow-green laser on Symphony cytometer

Results Summary

Good signal for PD-1 positive cells was obtained.
