I used this kit for my brain lysates (homogenized in RIPA buffer and purified with centrifugation). I detected 5-15 pg/mL in my sample wells (25-75 pg/mL in the original sample since the sample is diluted 5-fold). I was very pleased with the results since I had previously tried another ELISA kit which promised a 20 pg/mL detection and was unable to detect any of my samples. Overall, this kit was easy to understand and provided great results.

Tips

This product provides two separate vials for each item so that you can use the plates in two separate experiments. You would really limit the number of samples if you split it up more than that anyway. For brain lysates, you should perform a BCA analysis on your protein samples so you can normalize to pg/mg rather than pg/mL.

Results Summary

Each of my sample wells yielded a signal that could be quantified using the standard curve. I was pleased to find a difference between my LPS-treated mice and saline-treated mice. The difference was more slight than I would expect in cell culture experiments, but it was detectable with this ELISA kit.

Summary

The Good

The instructions are very easy to follow and optimized.

The Bad

They don't include the protocol sheets in the box anymore. This is to save paper which is nice, but I need to print it out anyway which is an inconvenience.

The Bottom Line

This is a sensitive ELISA that works well for lysates; I would buy it again if I repeat this type of experiment.
