To titrate the antibody, 2 x 10^6 splenocytes from C57BL/6 mouse were stained with: PerCP-Cy5.5 anti-CD4 Ab and Zombie NIR in 100 μL PBS at 4C for 30 min. Subsequently, the cells were fixed/permed using eBioscience™ Foxp3 / TF Staining Buffer Set according to manufacturer's instruction, then stained with: eF450 anti-FOXP3 Ab in 100 μL Perm buffer at 4C for 30 min.
