We are investigating the host innate immunity induced by bacterial infections. We used Pam3CSK4 as a positive control to examine the TLR2 signaling in our studies . Differentiated human macrophages will be treated with or without pam3CSK4 for 2 h and the activation of TLR2/NF-kB signaling will be evaluated by measuring the level of IL-6 in culture supernatants of macrophages.

As expected, treatment of human macrophages with 50 ng/ml Pam3CSK4 can significantly induce the production of IL-6 in human macrophages.
