To check the transfection efficiency of MEF cells, cells are 70–90% confluent at the time of transfection. Dilute 5 µl of Lipofectamine Reagent in 100 µl of Opti-MEM. Mix gently. Dilute 2.5 µg of YFP DNA in 100 µl of Opti-MEM. Mix gently. Incubate at room temperature for 5 minutes. Add diluted DNA to diluted Lipofectamine 2000 Reagent (1:1 ratio) and incubate at room temperature for 15 minutes. Add DNA-lipid complex to cells. Incubate at 37֯C incubator with 5% CO2. After 24 hour, take photos to check the transfection efficiency.
