These kits were used with SARS-CoV-2 convalescent sera samples as well as a negative control sera pool and a positive control sera pool. Our controls remained consistent across all replicates. When attempting to complete a high volume of assays using these kits, keep in mind there is an overnight incubation to begin with. Many reagents are presented in nearly exact volumes, which makes saving plate wells and reusing reagents nearly impossible. Overall, it does what it is supposed to do and is easy to use.

Tips

Plate rocker will suffice in absence of orbital shaker. Briefly vortex reagents before use. Do not forget to put lyophilized RBD Bridge Mix at -20C for storage.

Results Summary

This product helped us efficiently determine Ab levels (using optical density) of a ~75 sample cohort. When utilizing half the plate for a denaturing wash buffer, we were able to obtain avidity information as well. These data will be analyzed to observe any sex/race-based differences in Ab levels of our subjects.

The Bottom Line

Expensive kits, but they work very well. Depending on one's plate set up, there should be opportunities to remove wells and save them to be used for any repeat samples after the study, but the reagent amounts do not allow for this. This is a positive aspect as they are not wasting reagents, but several other kits provide extra reagents and that is what seems to be standard from my experience. Anywhere from 1-4 plates can be accurately managed and run by one operator, making this a very good option for high output studies.
