Our lab uses this antibody to test for Sox2 expression in cochlear cells (Sox2 is the pro sensory domain marker in the developing cochlea then gets restricted to supporting cells in the auditory region of the inner ear). It works well on whole tissue and sections. An antigen retrieval step prior to immunostaining (15min @ 80 deg C) worked wonders in terms of staining clarity!

Experimental Design and Results Summary

Applications

Immunohistochemistry

Sample

Mouse cochlea

Primary Incubation

1:250, primary incubation at 4 deg C overnight

Blocking Agent

10% normal goat serum

Secondary Incubation

1:500, 2 hours at room temperature on shaker

Tertiary Incubation

None

Detection

Fluorescence microscope/confocal LSM 780 488 and/or 594nm

Results Summary

This is a one week old wildtype mouse cochlea section. The SOX2 staining here marks all supporting cells and few greater epithelial ridge cells very faintly as expected.
