Human cervical cancer cells were stained with CD49f antibody and sorted for CD49f- positive and -negative cells. CD49f antibody is a cancer stem cell marker so positive cells were evaluated for tumorsphere growth.

Summary

The Good

Very easy to use.

The Bad

Optimization required for evaluation of expression.

The Bottom Line

CD49f can be co-stained with other cancer stem cell markers, including ALDEFLUOR. ALDEFLUOR must be performed at 37 degrees while CD49f must be done subsequently on ice to prevent efflux of ALDEFLUOR. Make sure to have cells stained only with CD49f as control for proper Flow Cytometry gating.
