Blocking binding of the antibody to unspecific proteins is an important step in immunocytochemistry which can achieved by using protein bovine serum albumin or skimmed milk. While performing immunofluorescence in cultured cells, sigma donkey serum serves as a very good blocker of non-specific reactions. 10% donkey serum in 0.2% TritonX-100 in PBS is a composition of blocking solution that works well for cultured cells when incubated for 2 hours at room temperature.
