It is well known that B lineage cells play a major role in pathogenesis of autoimmune diseases through presentation of antigen to T cells, production of autoantibodies and inflammatory cytokines that modulate immune responses. We are interested in elucidating the biology of B lineage cells during autoimmunity. In this experiment, I wanted to stain CD49d B cells.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Wild-type mouse spleen

Primary Incubation

Surface staining with CD49d PE-Vio 770 (1/800) CD19, etc. (20 min, 4° in the dark)

Blocking Agent

Clone 2.4G2 (Fc block)

Secondary Incubation

N/A

Tertiary Incubation

N/A

Detection

Flow cytometry, yelllow green laser

Results Summary

I could stain CD49d cells in the spleen using the antibody at 1/800. The histograms presented are gated in live CD19+ cells.
