The antibody can be used to analyse and purify the different stages of red blood cell differentiation in mouse models.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Mouse - Bone marrow and Fetal liver

Primary Incubation

1 microgram per mouse at 4' C for 60 min

Blocking Agent

PBS + 2 % FCS

Secondary Incubation

Not required

Tertiary Incubation

Not required

Detection

Flow cytometry

Results Summary

The antibody nicely separates the early and late stage progenitors of red blood cell differentiation in combination with TER119 antibody and can be effectively used to stain mouse embryonic foetal liver and adult mouse bone marrow. Particularly, the antibody signal is quite bright, therefore, antibody can also be used at a lower concentration and requires low voltage in FACS ARIA or Fortessa analyzer.

Summary

The Good

Extremely bright signal and clear separation of CD71 positive and negative cells.

The Bad

BV421 channel requires exposing the cells to UV LASER, which could be detrimental to the cells.

The Bottom Line

Highly recommend using this antibody, especially analysing and purifying red blood cell differentiation stages.
