Aim of the study was to test the effect of an on-target siRNA on CUL1 protein levels in human cells. The transfected cells were transfected with on-target siRNA, and after incubation for 36 hrs, the cultured cells were synchronized and treated with a growth factor. The cells were lysed, protein extracts were made and resolved in 10% SDS-PAGE. The resolved proteins were immunoblotted with CUL1 antibody to detect its levels. Selection of antibody was based on information provided in antibody flyer.
