The mouse tissues were homogenized, sonicated, and proteins were extracted in RIPA buffer. The extracted proteins were measured, and an equal amount of proteins were resolved in 7% SDS-PAGE. High range protein marker was used for this experiment. Resolved proteins were further immunoblotted for the detection of Cul3 protein using specific antibody obtained from Cell Signaling Technology. The selection of antibody was based on the merits shown in antibody flyer.

Enhanced chemiluminescent detection in X-Ray film
Although antibody had detected a few off-target non-specific epitopes, the specificity was high for its on-target Cul3.
Detected bright off-target epitopes of lower molecular weight
