I used this antibody to determine check the efficiency of p27 knockdown in 293T cells. scramble siRNA and p27 specific siRNA-transfected cells were subjected to western blot analysis.
Western Blot
Scramble siRNA and p27 specific siRNA-transfected 293T cells
1:1000 dilution, 18 hours at 4 degrees with gentle shaking
5% non-fat dry milk in Tris buffer containing 0.1% Tween-20
HRP-conjugated anti-rabbit antibody 1:2000 dilution, 1 hour at room temperature with gentle shaking

p27-specific siRNA did not work efficiently at knocking down the gene as depicted by the western blot.
